چکیده

Pyrimethamine is an antimalarial drug which possesses schizonticidal activity in both tissue and blood phases. Though pyrimethamine is known to inhibit folic acid metabolism of malarial parasites by binding with folio acid reductase (6), the cytological effect on the parasites is not completely understood. To our knowledge, there has been no study until now on the morphological effects of pyrimethamine on malarial parasites. However, Thurston (11), using light microscopy, reported the effects of a similar antimalarial drug, proguanil, on the nuclear division of the erythrocytic stages of Plasmodium berghei. Since proguanil and pyrimethamine exert qualitatively similar effects on malarial parasites (8), pyrimethamine is also thought to act against nuclear division of malarial parasites. In this investigation, we have studied with electron microscopy the morphological effects of pyrimethamine on the erythrocytic stages of a malarial parasite, Plasmodium gallinaceum. It is hoped that pyrimethamine may be used for the study of nuclear division in malarial parasites, which is still poorly understood.

Treatment of Malaria Parasitaemia in Infants and their Mothers

Malaria is an infection sustained by three parasites namely: Plasmodium falciparum, Plasmodium vivax, and Plasmodium ovale. Plasmodium falciparum is the most common and virulent parasite. These parasites are present in different areas of the sub-Saharan African countries and Asia. In 2010, there were an estimated 219 million cases of malaria resulting in 660,000 deaths and, approximately, two-t...

Rational drug design approach for overcoming drug resistance: application to pyrimethamine resistance in malaria.

Pyrimethamine acts by selectively inhibiting malarial dihydrofolate reductase-thymidylate synthase (DHFR-TS). Resistance in the most important human parasite, Plasmodium falciparum, initially results from an S108N mutation in the DHFR domain, with additional mutation (most commonly C59R or N51I or both) imparting much greater resistance. From a homology model of the 3-D structure of DHFR-TS, ra...

Prescription patterns and drug use among pregnant women with febrile Illnesses in Uganda: a survey in out-patient clinics

BACKGROUND Malaria is a public health problem in Uganda; affecting mainly women and children. Effective treatment has been hampered by over-diagnosis and over-treatment with anti-malarial drugs among patients presenting with fever. In order to understand the effect of drug pressure on sulfadoxine-pyrimethamine (SP) resistance in pregnancy, a sample of pregnant women presenting with fever in out...

Population genetics and drug resistance markers: an essential for malaria surveillance in Pakistan.

Plasmodium (P.) vivax is the prevalent malarial species accounting for 70% of malaria cases in Pakistan. However, baseline epidemiological data on P. vivax population structure and drug resistance are lacking from Pakistan. For population structure studies, molecular genetic markers, circumsporozoite protein (csp) and merozoite surface protein-1 (msp-1) are considered useful as these play an im...

Amodiaquine for treating malaria.

BACKGROUND Amodiaquine has been widely used to treat malaria. Due to reports of fatal adverse drug reactions, discontinuation or modification of its use has been suggested. OBJECTIVES The objective of this review was to assess the effects of amodiaquine for treating malaria. SEARCH STRATEGY We searched the Cochrane Infectious Diseases Group trials register and Medline. We also contacted res...
